Anti-SHISA3 antibody was prepared from whole rabbit serum produced by repeated immunizations with a 15 amino acid peptide near the internal region of human SHISA3.
Anti-SHISA3 Antibody was affinity purified from monospecific antiserum by immunoaffinity chromatography. SHISA3 antibody is predicted to not cross-react with other SHISA protein family members.

Anti-SHISA3 Antibody has been tested for use in ELISA, Western Blotting, Immunohistochemistry and Immunofluorescence. Specific conditions for reactivity should be optimized by the end user. Expect a band at approximately 26 kDa in Western Blots of specific cell lysates and tissues.

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

SHISA3 plays an essential role in the maturation of presomitic mesoderm cells by individual attenuation of both FGF and WNT signaling. The Shisa family of single-transmembrane proteins is characterized by an N-terminal cysteine-rich domain and a proline-rich C-terminal region. Its founding member, Xenopus Shisa, promotes head development by antagonizing Wnt and FGF signaling. Shisa physically interacted with immature forms of the Wnt receptor Frizzled and the FGF receptor within the ER and inhibited their posttranslational maturation and trafficking to the cell surface. Loss of Shisa function sensitized the neuroectoderm to Wnt signaling and suppressed head formation during gastrulation.
