Anti-PD-1 antibody was prepared from whole rabbit serum produced by repeated immunizations with a 16 amino acid synthetic peptide from near the internal region of human PD-1.
Anti-PD-1 Antibody was affinity purified from monospecific antiserum by immunoaffinity chromatography. Cross reactivity with PD-1 from other sources has not been determined.

Anti-PD-1 Antibody has been tested for use in ELISA, Western Blotting, Immunofluorescence, and Immunohistochemistry. Expect a band at approximately 32 kDa in Western Blots using positive control THP-1 whole cell lysate, Human brain tissue for IHC, or other specific cell lysates and tissues. Specific conditions for reactivity should be optimized by the end user.

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.
Expiration date is one (1) year from date of receipt.
Cell-mediated immune responses are initiated by T lymphocytes that are themselves stimulated by cognate peptides bound to MHC molecules on antigen-presenting cells (APC). T-cell activation is generally self-limited as activated T cells express receptors such as PD-1 (also known as PDCD-1) that mediate inhibitory signals from the APC. PD-1 can bind two different but related ligands, PDL-1 and PDL-2. Upon binding to either of these ligands, signals generated by PD-1 inhibit the activation of the immune response in the absence of "danger signals" such as LPS or other molecules associated with bacteria or other pathogens. Evidence for this is seen in PD1-null mice who exhibit hyperactivated immune systems and autoimmune diseases.
