Conjugated Peptide
This monoclonal antibody was produced by repeated immunizations with a synthetic peptide corresponding to the carboxy terminus of human Ki-67 protein. The hybridoma was produced by fusing New Zealand White rabbit splenocytes and myeloma cells using conventional technology.
This product is clarified tissue culture supernatant. The antibody is specific for human Ki-67 protein. Cross-reactivity occurs with rat Ki-67. Cross reactivity with Ki-67 protein from other sources has not been determined.
Application Details
IHC
This monoclonal antibody contains TBS/1% BSA. Anti-Ki-67 has been tested with IHC and is suitable for ELISA and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 345 and 395 kDa in size corresponding to Ki-67 protein by western blotting in the appropriate cell lysate or extract. For immunohistochemistry, samples should be formalin fixed and paraffin embedded. Deparaffinize slides using xylene or xylene alternatives and graded alcohols. Staining requires boiling of sections in 10 mM citrate buffer pH 6.0 for 10 min followed by cooling at RT for 20 min. Ki-67 is a nuclear protein. Tonsil tissue can be used as a positive control.

The MKI67 gene encodes a nuclear protein termed Ki-67 that is associated with and may be necessary for cellular proliferation. Alternatively spliced transcript variants have been described. A related pseudogene exists on chromosome X. Two isoforms have been reported, with isoform 1 transcribed as the longer transcript. Ki-67 is a nuclear protein predominantly localized in the G1 phase in the perinucleolar region. In later phases it is also detected throughout the nuclear interior, being predominantly localized in the nuclear matrix. In mitosis, it is present on all chromosomes.
