Application Details

ELISA
Useful in Sandwich ELISA for Quantitative Detection of Antigen. Aliquot 0.1ml per well of the 2000pg/ml,1000pg/ml, 500pg/ml, 250pg/ml, 125pg/ml, 62.5pg/ml, 31.2pg/ml rat FAS-L standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of rat cell culture supernates, serum or plasma(heparin, EDTA, citrate) to each empty well. It is recommended that each rat FAS-L standard solution and each sample be measured in duplicate.

Store vials at 4°C prior to opening. Centrifuge product if not completely clear after standing at room temperature. This product is stable for 6 months at 4°C as an undiluted liquid. Dilute only prior to immediate use. For extended storage freeze at -20°C or below for 12 months. Avoid cycles of freezing and thawing.

FAS Ligand (FASL) is a 40 kDa type II membrane protein belonging to the tumor necrosis factor family, which induces apoptosis by binding to its receptor, Fas. The human FasL gene consists of approximately 8.0 kb and is split into four exons. This gene consists of 281 amino acids with a calculated M(r) of 31,759 and was mapped on chromosome 1q23. It has an identity of 76.9% at the amino acid sequence level with mouse FasL. The FAS and FASL system plays a key role in regulating apoptotic cell death and corruption of this signaling pathway has been shown to participate in immune escape and tumor genesis. FAS and FASL triggered apoptosis pathway plays an important role in human carcinogenesis. This system may also play a role in modulating the genetic susceptibility of mouse strains to develop T-cell lymphoblastic lymphomas.
