Biodistribution of 2'MOE-Modified ASOs
Evaluating tissue distribution of MOE-modified ONTs is critical for assessing delivery efficiency, off-target accumulation, and pharmacological activity. Reliable detection in complex tissues guides preclinical candidate selection and supports translational development.
In mouse liver tissue, Anti-MOE antibody clone MOE4 (green) detected accumulation of MOE-modified ASOs in non-parenchymal cells surrounding hepatocytes. No staining was observed in PBS-treated controls, confirming high specificity. These results demonstrate the reproducibility of ModDetect™ antibodies in tissue biodistribution workflows.
Combined Detection of PS and 2-MOE Modifications
Oligonucleotide therapeutics often incorporate multiple chemical modifications to balance stability, uptake, and activity. ModDetect™ Anti-PS (clone PS03) and Anti-MOE (clone MOE4) antibodies were used together to detect a 2′-MOE-modified ASO 5-10-5 gapmer containing ten PS bonds following subcutaneous delivery in mice (50 mg/kg, 72 h).
Both antibodies produced strong, specific staining patterns consistent with the expected biodistribution of the modified ASO, confirming the compatibility of ModDetect reagents for dual-modified oligonucleotide analysis in vivo.
Co-Localization of MOE-Modified ASOs
Co-localization studies are essential for confirming that MOE-modified ONTs are properly internalized and trafficked. Overlap between therapeutic oligonucleotides and Anti-MOE staining validates antibody specificity and provides mechanistic insights into cellular uptake.
In HeLa cells, Cy5-conjugated PS-MOE ASOs (magenta) were co-detected with Anti-MOE antibody clone MOE-C (red). Strong overlap confirmed faithful recognition of MOE modifications and alignment with therapeutic ASO signals. These results validate the use of ModDetect™ Anti-MOE antibodies for subcellular localization studies.
